The Melanocortin Receptor Family: A Guide for Research

The melanocortin system is a masterclass in receptor biology: one parent hormone family signaling through five receptor subtypes, each governing different physiology. Four research compounds in our catalog map onto this system, and knowing which engages which receptor is essential for clean study design.

The Receptor Map

MC1R governs melanin synthesis in melanocytes. MC2R responds to ACTH in the adrenal cortex. MC3R and MC4R are expressed centrally and dominate energy balance and behavioral research. MC5R appears in exocrine function studies.

The Research Compounds

Melanotan 1 is the selective probe โ€” a linear alpha-MSH analog with strong MC1R preference, suited to pigmentation-focused designs. Melanotan 2 is the broad probe โ€” a cyclic analog engaging MC1R, MC3R, and MC4R, useful when mapping cross-receptor effects. PT-141, derived from Melanotan 2, centers on MC4R and anchors central melanocortin research. KPV, the C-terminal tripeptide of alpha-MSH, is studied in inflammation signaling without pigmentary activity.
