Mitochondrial biology sits at the center of modern aging-pathway research, and three compounds appear constantly in the literature: MOTS-c, SS-31, and NAD+. They are often grouped together, but each operates at a different level of mitochondrial function — which is precisely what makes them complementary research tools.
MOTS-c: The Mitochondrial-Derived Peptide
MOTS-c is a 16 amino acid peptide encoded within the mitochondrial genome itself โ one of the founding members of the mitochondrial-derived peptide class. Research examines its activation of the AMPK pathway, its role in glucose and fat metabolism, and its behavior as a mitochondria-to-nucleus signaling molecule in exercise physiology and metabolic studies.
SS-31: The Membrane Stabilizer
SS-31, of the elamipretide class, takes a structural approach. This Szeto-Schiller tetrapeptide concentrates in the inner mitochondrial membrane and binds cardiolipin, the phospholipid that organizes the electron transport chain. Preclinical studies examine whether stabilizing this architecture preserves ATP production and reduces reactive oxygen species in models of cardiac, renal, and muscular dysfunction.
NAD+: The Substrate Itself
NAD+ is not a peptide at all but the central redox coenzyme of cellular energy metabolism โ required for ATP production and consumed by sirtuins and PARP enzymes. Because tissue NAD+ declines with age, it anchors an enormous body of longevity-pathway research spanning metabolism, DNA repair signaling, and mitochondrial biogenesis.
Three Levels, One System
A useful mental model from the literature: NAD+ is the fuel-side substrate, SS-31 protects the machinery, and MOTS-c is the signal the machinery sends back to the cell. Laboratories designing mitochondrial studies often select compounds across these levels depending on whether the research question concerns substrate availability, structural integrity, or retrograde signaling.
